In a female patient (IPD2) with congenital myasthenic syndrome-22 (CMS22\; 616224), Regal et al. (2018) identified compound heterozygosity for mutations in the PREPL gene. The maternal allele carried a 4-bp deletion in exon 3 (c.358_361delTTTG, NM_001171603.1), resulting in frameshift and premature termination of the protein (Val121IlefsTer121). The paternal allele carried a 6-kb intragenic deletion
