Abstract :
A ligand fishing strategy was developed to screen for pancreatic lipase (PL) inhibitory peptides from quinoa protein hydrolysate (QPH) using PL-immobilized FeO@SiO magnetic particles. An initial activity of 230 U/g, a PL loading capacity of 180 mg/g, an immobilization yield of 25 %, and retained 39.23 % of its activity after 10 reuse cycles. Three novel peptides (LNEILAHE, VETELEKR, TVSAPSPK) were identified by LC-MS/MS and exhibited moderate PL inhibitory activities with IC50 values ranging from 0.48 to 0.92 mg/mL. They showed dose-dependent anti-adipogenesis effects in 3T3-L1 cells, with TVSAPSPK reducing intracellular TG content by 66.36 % at 1500 mg/mL. Kinetic analysis revealed that LNEILAHE and TVSAPSPK acted as mixed-competitive inhibitors, while VETELEKR showed anti-competitive inhibition. Molecular docking indicated that these peptides bound to the PL active site via hydrogen bonding, electrostatic and hydrophobic interactions. These findings validate the ligand-fishing strategy for peptide screening and highlight QPH-derived peptides as promising anti-obesity candidates. |
