Covalent conjugation of the natural monoterpene perillyl alcohol (POH) to the alkylating agent temozolomide (TMZ) (NEO100, highly purified perillyl alcohol). The half-life of NEO212 in mouse plasma was 94 min. Oral TMZ is the current chemotherapeutic standard of care for the treatment of patients with newly diagnosed glioblastoma. POH, a naturally occurring monoterpene and metabolic product of limonene. It is commonly found in the essential oils of several plants and fruits, such as peppermint, spearmint, cherries, and celery seeds. POH has long been reported to have anticancer effects in vitro and in vivo
