Homo sapiens (Human) dipeptidyl peptidase 9 DPP9 DPRP2
DPP8 and DPP9 are cytosolic peptidases. Have dpp activity similar to dpp4. The first identified endogenous DPP9 substrate is the tumor epitope RU1(34-42) (VPYGSFKHV). DPP9 is important in the immune system, DNA repair and mitochondria homeostasis. Inhibition or silencing of DPP9, but not DPP8, led to increased presentation of this antigen on MHC class I alleles to cytotoxic T-cells, linking DPP9 to the MHC class I antigen presentation pathway. Small ubiquitin-like protein modifier SUMO1 acts as an allosteric activator of DPP9. By binding to an armlike motif in DPP9\; DPP9 functions as an endogenous inhibitor of NLRP1 inflammasome. DPP9 interacts with a unique autoproteolytic domain (Function to Find Domain (FIIND)) found in NLRP1 and CARD8. DPP9 acts as a molecular switch that regulates key proteins, such as the tumor-suppressor BRCA2. Hatipoglu immunodeficiency syndrome\; HATIS. The symptoms associated with this monogenic disease partially overlapped with those of AIADK caused by an NLRP1 gain-of-function mutation. These included failure to thrive (consisting of poor weight gain,growing below the third percentile),skin manifestations, pancytopenia, and susceptibility to infections, which required hematopoietic stem cell transplant. The stop-gain mutations were expected to completely abolish the enzymatic activity of DPP9.
